アメリカの内科医ザイネ・カイムは、一九八〇年に出版した著書『日光があなたの生命を救う(Sun light Could Save Your Life)』(邦訳未刊)のなかで、四一歳の乳がん患者に日光療法を行なった例を紹介している。この患者はすでに乳房を切除し、化学療法も受けていたが効果はなく、がんが肺と骨に転移していた。カイムはがん自体を治療するのではなく、全身の健康状態を改善することを試みた。口にするのは未精製・未加工の食べ物だけで、多価不飽和脂肪酸を多く含む油脂は徹底的に排除する。そして日光浴を積極的に行なった。すると患者はわずか数か月で仕事に復帰し、転移したがんの症状も現われなかった。
The FDA has recently received inquiries regarding the presence of human papillomavirus (HPV) DNA fragments in Gardasil and is aware that information related to this issue is on the internet. A concern that the presence of these fragments could represent contamination of the vaccine arises from an unpublished report that recombinant HPV L1-specific DNA sequences were present in 13 vials of Gardasil from different lots.
The presence of DNA fragments is expected in Gardasil and not evidence of contamination. Based on the scientific information available to FDA, Gardasil continues to be safe and effective, and its benefits continue to outweigh its risks.
For Gardasil vaccine production, the HPV DNA encoding the L1 capsid proteins is inserted into the plasmid pGAL110 to transform yeast cells for production of VLPs [5,67,68]. Possible expression systems also include mammalian cells . Retention of residual recombinant DNA in protein-based vaccines has been a concern in the industry since induction of cancer is a single-cell phenomenon, and a single functional unit of foreign DNA integrated into the host cell genome might serve to induce cell transformation as a single event or part of a series of multifactorial events . Chromosomal integration of foreign DNA may occur through poorly understood mechanisms [71,72] with uncertain consequences . The short-term and long-term impact of the residual fragments of HPV L1 gene DNA or plasmid DNA if chemically bound to the mineral aluminum of AAHS nanoparticles is largely unknown and warrants further investigation.
DNA has the unique ability to integrate stably into the genome or to be maintained long term in an episomal form. This enables prolonged expression of the antigen of interest and enhancement of immunologic memory
cells with HPV integration usually exhibit deletion of L1, rendering these cells ineffective as a target for HPV L1-specific CD8+ T cells.
DNA present in the vaccine may integrate into the host genome
Fortunately, it is estimated that the frequency of integration is much lower than that of spontaneous mutation and integration should not pose any real risk
Progression is frequently associated with conversion of the viral genome from an episomal form to an integrated form, along with deletion or inactivation of E2, a negative regulator of E6 and E7 expression
Capsid proteins L1 and L2 attach to epithelial cell receptors and a long process of entry commences, resulting in cytoplasmic uncoating of the virus and entry of its genome into the nucleus of the infected cell, where it is transcribed and then replicated. Early proteins are expressed first and regulate the host cell life cycle and genome replication. They also regulate the expression of late proteins in a cell differentiation-dependent manner:
